Background
R-Spondin 1 (RSPO1) is a secreted growth factor that potentiates canonical Wnt/β-catenin signaling by binding to LGR4/5/6 receptors and neutralizing the activity of the E3 ubiquitin ligases RNF43 and ZNRF3. Through amplification of Wnt signaling, RSPO1 plays essential roles in stem cell maintenance, epithelial cell proliferation, tissue regeneration, and developmental processes. RSPO1 is widely used in cell biology and organoid research, particularly for the culture and expansion of intestinal and epithelial stem cells, and is also implicated in cancer-related Wnt signaling dysregulation.
