Background
R-Spondin 3 (RSPO3) is a secreted growth factor that enhances canonical Wnt/β-catenin signaling through interaction with LGR4/5/6 receptors and inhibition of RNF43/ZNRF3-mediated receptor turnover. RSPO3 plays essential roles in stem cell maintenance, angiogenesis, vascular development, tissue regeneration, and embryonic morphogenesis. It is widely used in organoid culture systems to support stem cell proliferation and epithelial growth. Aberrant RSPO3-mediated Wnt signaling has also been implicated in cancer progression and tumor microenvironment regulation.
