TEV Protease is a recombinant form of nuclear inclusion a (NIa) protease from tobacco etch virus (TEV). It has often been used as a biotechnological tool for cleavage affinity purification tags such as maltose-binding protein (MBP) or poly-histidine from fusion proteins. TEV Protease specifically recognizes the cleavage site: Glu-Asn-Leu-Tyr-Phe-Gln↓Gly/Ser (ENLYFQ↓G/S). TEV protease has a His-tag for easy removal from a cleavage reaction by using nickel affinity resins.
Leadgene® TEV Protease ELISA kit is an enzyme-linked immunosorbent assay (ELISA) for the quantitative detection of TEV Protease level in cleavage sample solution. The TEV Protease ELISA kit is for research use only (RUO). Not suitable for use in diagnostic or therapeutic procedures.

- Leadgene TEV Protease ELISA Kit is designed for commonly used mutant TEV. We have incorporated one modification (S219V) on the antigen used for ELISA antibody production.
- The S219V mutation in the TEV protease catalytic domain increases its stability and efficiency, making it highly resistant to autolysis and preventing autoinactivation, thus ideal for applications requiring protease stability.
Testprinzip
TEV Protease ELISA kit is used to detect TEV Protease in samples by sandwich ELISA method. This assay uses microplate pre-coated with mouse anti-TEV Protease monoclonal antibody to the solid phase. TEV Protease in the samples conjugates on solid phase and then react with the HRP conjugated mouse anti-TEV Protease monoclonal antibody. Subsequent wash steps will residual unbound antibody. After incubation with substrate solution, the reaction is determined by the absorbance at 450 nm. Quantification of TEV Protease level is accomplished by comparing the absorbance with standard curve.
