Lateral flow

Lateral flow assay development service

Lateral flow assay, also called lateral flow immunochromatographic assay, detects the targeting molecules in a liquid sample without an expansive instrument. This method is rapid and low-cost and, widely used in medical diagnostics.

The principle of lateral flow assay is the usage of the colloidal gold (Au) nanoparticle-labeled antibody on the conjugate pad. The Au labeled antibody binds to the targeting antigen/antibody and shows purple color.

   

 

There are several lateral flow assays on the detection of antigen or antibody:

  •  Detection of antigen in the liquid sample

The antigens are applied to the sample pad and bind to the Au labeled antibodies on the conjugate pad. When the Au-antibody-antigen complex flows through the test line (T line), the complex is captured by the secondary antibody, and the Au nanoparticles show purple color. Unbound antibodies continue to flow through the control line (C line.) The third antibodies on the T line recognize the Au labeled antibodies and show color to ensure the completion of the test.

 

 

  • Detection of antibody in the liquid sample

The antibodies are applied to the sample pad and bind to the Au labeled antigens on the conjugate pad. When the Au-antibody-antigen complex flows through the test line (T line), the complex is captured by the secondary antibody, and the Au nanoparticles show purple color. In the meanwhile, the Au labeled antibodies on the conjugate pad also flow along with the sample. These antibodies will be captured by the antibodies immobilized to the control line (C line,) and the C line will show the purple color to ensure the completion of the test.
